AB OBJECTIVE: Errors in addition of sample, manually or with automatic
dispensers, affect the sensitivity of assays. A sample diluent was
therefore developed, which would confirm that sample had been added to
the well, thereby confirming that the protocol was followed. RESULTS:
The diluent is green/brown and changes to blue when serum or plasma is
added to it. This change is easily seen and can be recorded
spectrophotometrically (absorbance increases from 0.05 to 0.35). Assay
sensitivity has been improved with the following bleeds detected of BBI
HIV-1 seroconversion panels; BBI R3, L2, Q5, I2, S2 and of NABI panels;
151-2C, 171-2 C and 121-2C. The sensitivity of HIV-2 detection has also
been increased. CONCLUSION: The inclusion of a Sample Addition Monitor
facilitates the confirmation that sample has been added to the well.
Mistakes can be easily identified and rectified thereby improving the
safety of blood supply.
